I used this kit to isolate mouse CD4+ T-cells from spleen. This bead based column isolation, helps me to use both positive and negative selection fractions for analysis.

All that you need to do is just follow the protocol from Miltenyi as such. Always count the initial number of cells taken for isolation. Don't vary with antibody cock tail to be used and number of washes recommended as it may affect final purity.
Protocol is straight forward and very simple and just follow it.
I always get more than 90% pure CD4+ T-cell population

Purity is good and it you can use both CD4+ and CD4- populations after purification
Expensive and need to purchase column separately.
