Product Name:

PE Anti-Human CD11b (ICRF44)

Catalog Number:

50-0118

HL-60 cells are able to differentiate to neutrophils after all trans retinoic acid (RA) treatment. They are also able to differentiate to monocytes after 1,25-dihydroxyvitamin D3 (D3) treatment. A characteristic of this conversion is CD11b expression. Cell culture volumes containing one million cells (untreated vs. RA treated) were spun down and resuspended in 200µl PBS with 2.5µl CD11b Tonbo 50-0118 antibody. This antibody has high specificity and is at a competitive price compared to similar antibodies.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

HL-60 cells ; untreated, all trans retinoic acid (RA), 1,25-dihydroxyvitamin D3 (D3) treated

Primary Incubation

1 million cells were incubated 1h , 37C, in 200µl PBS with 2.5µl CD11b antibody

Blocking Agent

None

Secondary Incubation

None

Tertiary Incubation

None

Detection

Instrument: BD LSRII. PE was excited at 488 nm and emission was collected through 550 long-pass dichroic and 576/26 band-pass filters.

Results Summary

The PE Anti-Human CD11b (ICRF44) has some background. This makes small changes in expression hard to detect (in our case RA vs C). Samples with high expression (D3 in our case) are still acceptably labeled.
