Our lab digests large amounts of both human and animal tissue for single cell isolation. This machine very quickly dissociates many types of tissue that cannot digest in enzyme buffer alone. We use it for lung, spleen, intestine, and lymph nodes.

Protocol Overview

After chopping whole human tissue into smaller chunks, we digest the tissue in collagenase buffer. If mouse tissue, we place in whole. Then we use the pre-set tissue setting 1-2 times depending on the tissue and cells are completely dissociated. Using this machine only takes a minute or two.

Tips

Make sure C-tubes are secure in the slots before operation.

Results Summary

We are able to get single cells from spleen, lung, lymph nodes, and intestine using this separated for downstream analysis such as flow cytometry.
