This review is about L-selectin staining on mouse lymphocytes. In the mouse, L-selectin is detected on most thymocytes, with the highest levels of expression on an immunocompetent subset and a population of dividing progenitor cells, and on peripheral leukocytes, including subsets of B and T lymphocytes, neutrophils, monocytes, and eosinophils. L-selectin is rapidly shed from lymphocytes and neutrophils upon cellular activation, which is what I wanted to test. It can also be used for differentiating effector vs central memory T cells within CD44high subset.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Mouse splenocytes

Primary Incubation

25 minutes on ice

Blocking Agent

CD16/32 antibody for blocking Fc receptors

Secondary Incubation

None

Tertiary Incubation

None

Detection

Flow cytometry on BV650 channel

Results Summary

The staining showed that most cells were positive for CD62L. Within the activated cells, CD62L was down-regulated, which was expected and consistent with literature.

Summary

The Good

The staining was very bright and relevant

The Bad
