Type-2 diabetic rats were sacrificed at 6 weeks of treatments and kidney was harvested in sterile PBS. Kidney tissues were homogenized, protein extract were prepared, and resolved in 10% SDS-PAGE. Resolved proteins were then immunoblotted with phospho-MST1 (Thr183)/MST2 primary antibody and HRP-conjugated anti-species secondary antibody. Selection of antibody was done based on information provided in antibody flyers.
Western Blot
Rat tissue protein lysate
Overnight at 4 0C (5% BSA in TBS with 0.1% Tween 20; dilution 1:1500)
5 % Skimmed milk in TBS with 0.1% Tween 20 (for 1 hr at RT)
1 hr at RT (5% skimmed milk in TBS with 0.1% Tween 20; dilution 1:4000)

Good quality protein bands of phospho-MST1 (Thr183)/MST2 were detected in samples. All bands were very clear, sharp and bright. There was no non-specific signals were detected.
