In the present study, liver, kidney from diabetic and non diabetic rats were harvested and tested for cofilin protein levels. In brief, tussue were homogenized in RIPA buffer, and centrifuged at 10000 xg for 10 minutes. Supernatant was collected and proteins were estimated using micro BCA method. Further, an equal amount of proteins were resolved in 12% SDS PAGE and immunoblotted with cofilin antibody

Overnight at 4 0C (5% skimmed milk in TBS with 0.1% Tween 20; dilution 1:2000)
5 % Skimmed milk in TBS with 0.1% Tween 20 (for 1 hr at RT)
1 hr at RT (5% skimmed milk in TBS with 0.1% Tween 20; dilution 1:5000)
