In the present study, rat hepatocytes were isolated using standard protocol, in vitro cell culture were established, stained with cell tracker green and assessed for cell migration assay.
Staining of rat hepatocyte for in vitro migration assay
Rat pancreatic cell culture, DMEM, DMEM with 10% FBS, PBS, DMSO, culture incubator, fluorescent microscope
Five rats of 12 weeks age were sacrificed, livers were harvested, ectracellular matrix of liver tissue was digested with collagenase V and isolated cells are then filtered through a 100 µm pore size mesh nylon filter, and cultured onto plates. After 4-hour culture, hepatocytes were selectively purified using asialoglycoprotein receptor (ASGPR) antibody conjugated dynabeads. Established culture was passaged for at least 2 times and stained with 4 micromolar solution of Cell Tracker Green CMFDA dye for 40 min. After staining media was replaced with DMEM containing 10% serum and cells were used for in vitro cell migration assay.
