Aim of the present study was to detect cytoskeleton protein vinculin levels in rat muscle tissue. In brief, muscle tissue from rats of all treatments were used to make protein lysate and resolved in polyacrylamide gel. Further, resolved proteins were immunoblotted for vinculin protein and after ECL treatment blots were developed in x-ray film.
Western Blot
Rat whole muscle tissue protein lysate
Overnight at 4 0C (5% skimmed milk in TBS with 0.1% Tween 20; dilution 1:1000)
5 % Skimmed milk in TBS with 0.1% Tween 20 (for 1 hr at RT)
1 hr at RT (5% skimmed milk in TBS with 0.1% Tween 20; dilution 1:5000)

A bright and strong signal of vinculin protein at around 120 KDa was observed in rat samples. However, no or very faint band was observed in control rat samples.
A very bright band of vinculin was observed
Lot of non specific bands of low molecular weight proteins were observed
Overall best quality anti-vinculin antibody to detect vinculin proteins in rat tissue samples.
