Starting Material

Rat pancreatic whole tissue lysate, SDS-PAGE assembly, immunoblotting assembly, x-Ray films, autoradiography cassette, x-ray film developer and fixer etc.

Protocol Overview

Diabetic and non-diabetic rats were sacrificed following standard ethical conditions. Pancreatic tissues from rats were harvested and whole tissue extract were prepared in protein extraction buffer. Further, an equal amount (20 micro gram) of proteins were resolved in polyacrylamide gel electrophoresis and western transferred to PVDF membrane. Membrane were probed for cdc24 and c-Raf proteins and treated with HRP conjugated secondary antibody. Probed blots were ECL developed with Supersignal west dura extended duration substrate for 30 seconds and signals were captured in x-Ray film
