Aim of the present study was to mount whole pancreatic tissue stained with Texas red conjugated isolectin B4 in artificially induced diabetic rats fed with or without a number of varieties of milk samples.

Protocol Overview

Diabetic rats under specific treatments were sacrificed and pancreas were isolated. Whole pancreas were fixed in 4% paraformaldehyde for 1h, washed times with PBS and permeabilized with 0.5% triton X100. Permeabilized tissue were incubated with isolectin-B4 solution for overnight and washed 4 times for 15 min each with cold PBS. Stained pancreatic tissue were mounted on microscopic slide using Vectashield Hardset Mounting medium and vasculature were observed under fluorescent microscope. Slides were kept in a dark place at room temperature so that mounting medium will set hard.
