Aim of the present study to detect PROX1 surface proteins in retinal tissue sections. For this, rats were sacrificed, eyes were enucleated, fixed in 4% paraformaldehyde and retinal cryosection were made. Retinal cryosection were hydrated with PBS, blocked with 3% BSA and probed with PROX1 antibody obtained from Abacm. Slides were examined in a fluorescent microscope with 400 x optical magnification. Selection of antibody was based on previous experience.
Immunofluorescence
Rat retinal sections
Overnight at 4 0C (3% BSA in PBS; dilution 1:100)
3 % BSA in PBS containing 0.5% normal goat serum (for 1 hr at RT)
1 hr at RT (3% BSA; dilution 1:500)

Alexa fluor conjugated secondary antibody
PROX1 antibody from abcam has given very high specific immunostaining of PROX1 proteins in rat retinal section. No non specific binding of PROX1 antibody were observed in rat samples
Antibody is highly specific to PROX1 in rat proteins
Overall highly specific of Ancam to detect PROX1 in rat samples.
