In the present study pPAK1 + pPAK2 levels was measured in muscles of rats subjected to hind limb ischemia and normal levels of PAK1 were also measured. In brief, femoral artery of male albino rats of western strain wister were ligated and cut and after 7 days femoral muscle were harvested. After that, ischemic muscle proteins were extracted, resolved in SDS-PAGE and immunoblotted with anti-PAK1 antibody for PAK1 levels. Antibody selection was based on information’s given in antibody flyers.
Western Blot
Whole rat tissue protein lysate
Overnight at 4 0C (5% skimmed milk in TBS with 0.1% Tween 20; dilution 1:1500)
5 % Skimmed milk in TBS with 0.1% Tween 20 (for 2 hr at RT)
2 hr at RT (5% skimmed milk in TBS with 0.1% Tween 20; dilution 1:4000)

PAK1 antibody from abcam has given very specific and intense band of Pak1 at correct size. However, there was a non specific band with low intensity at higher molecular weight.
Antibody is highly specific to PAK1 proteins in rat proteins
A non specific band was also observed at 78 KDa
Good specificity of PAK1 antibody.
