In the present study male albino rats of two months age were subjected to hind limb ischemia. After recovery, rats were kept on standard nutritional condition with different phytoextract treatments. After specific time period, rats were sacrificed, femoral muscles were harvested an equal amount of extracted proteins were resolved in SDS-PAGE and immunoblotted for pSTAT3 levels. Selection of antibody was based on information provided in antibody flyer.

Overnight at 4 0C (5% BSA in TBS with 0.1% Tween 20; dilution 1:500)
5 % Skimmed milk in TBS with 0.1% Tween 20 (for 2 hr at RT)
1 hr at RT (5% skimmed milk in TBS with 0.1% Tween 20; dilution 1:4000)

Bright, intense and highly specific bands of pSTAT3 were detected in rat protein samples. However, one non specific band with very low specificity was also observed.
