Male albino rats kept at specific treatments were sacrificed and retinas were harvested. Retinal protein extract were prepared in RIPA buffer and equal quantities of proteins were resolved in 8% SDS-PAGE. Resolved proteins were western transferred in nitrocellulose membrane and immunoblotted for NLRP3 protein levels. Abcam antibody was selected because of information provided in antibody flyer and previous experience of antibody for reactivity with human protein.

Rat retina tissue whole protein lysate
Overnight at 4 0C (5% Skimmed milk in TBS with 0.1% Tween 20; dilution 1:1000)
5 % Skimmed milk in TBS with 0.1% Tween 20 (for 2hr at RT)
1hr at RT (5% skimmed milk in TBS with 0.1% Tween 20; dilution 1:5000)

Lot of bright non specific bands was observed. NRLP3 specific band at 120KDa was of weak intensity and were merged with other non specific bands. This indicated poor specificity and reactivity of abcam antibody with rat proteins.
