The aim of the experiment was to estimate the level of glutathione in rat liver tissue. Rat liver tissue was weighed, cleaned with phosphate buffer saline, homogenized in cold 50 mM phosphate buffer and centrifuged. The supernatant obtained was deproteinated and assayed for the presence of glutathione by Glutathione Assay Kit. To the 50 microlitre of the sample, 150 microlitre of assay cocktail (containing MES buffer, cofactor mixture, enzyme mixture, water and DTNB). The Assay was based on the kinetic method and absorbance was read at 410 nm. This kit gives good results and reproducibility was observed.

Rat liver tissue was rinsed with phosphate buffer saline and then homogenized in homogenizer with an appropriate buffer. The homogenized sample was centrifuged to remove the debris and supernatant was collected. Any interfering protein was also removed. The glutathione level in the supernatant was estimated using the Glutathione Assay Kit according to the manufacturer’s instructions. Absorbance was taken at 410 nm. The amount of glutathione in sample was estimated by using standard curve of glutathione.
