RayBio® Rat Insulin ELISA Kit  is sandwich ELISA based kit and used to detect the presence of insulin in plasma. Blood from rat’s orbital venous plexus was collected in EDTA coated tubes. Blood was centrifuged to separate the plasma from red blood cells. Rat plasma insulin levels were measured by using Rat Insulin ELISA kit. Antibody coated plates were incubated with sample (plasma), incubated and washed to remove any unbound antibody. Further, biotinylated anti-rat insulin antibody is added, washed and then incubated with TMB substrate. In the presence of insulin bound biotinylated anti-rat insulin antibody, TMB is converted to the coloured product which was read at 450nm.

Protocol Overview

Blood collected in EDTA coated vaccutainers from rats. The blood was centrifuged and the upper portion (plasma) was collected. The insulin level was estimated in plasma by using RayBio® Rat Insulin ELISA Kit. All the steps were performed according to the manufacturer’s instruction. Readings were taken at 450 nm and insulin level was estimated by using standard curve.
