HL-60 cells are able to differentiate to neutrophils after all trans retinoic acid treatment. A characteristic of this conversion is CD38 expression. We treated HL-60 cells with all trans retinoic acid 0.1 µM final concentration for 48h. Cell culture volumes containing one million cells (untreated vs. RA treated) were spun down and resuspended in 200µl PBS with 2.5µl CD38 antibody. CD38 antibodies from three companies were compared. BD PE Mouse Anti-Human CD38 Clone HIT2 (RUO) has very low background, good resolution, and an affordable price.
