Our lab does a lot of cell culture with cell lines as well as primary human cells. Because it is crucial to avoid mycoplasma contamination of precious patient samples we routinely test for mycoplasma contamination of our cultures. We chose this kit because of straightforward protocol and fast results.

Starting Material

Media of cell culture (different cell lines and primary cell cultures)

Protocol Overview

Collect some media (with some cells) from the culture you would like to test. Heat samples at 100C for 15min. Add 50uL of the heated sample to a well of a 96 well plate. Use 50uL of positive control and negative control in separate wells. Prepare the HEK-Blue cell suspension (according to protocol) and add 200uL of cell suspension in each well. Incubate plate in a CO2 incubator over night. Wells that turn purple are mycoplasma positive, wells remaining pink are mycoplasma negative.
