Our group studies T cells in the blood of untreated chronic HIV+ individuals. We are constantly expanding and optimizing our T cell panels to measure various T cell subsets, activation, and inhibitory receptor phenotype. Occasionally we study B cells as well, and CD86-PE has worked well in our B-cell-focused panel. We chose CD86-PE because it fit well in a 7-color panel characterizing B cell activation.

Results Summary

1uL CD86-PE works great to detect B cell activation. Cells were incubated with or without 1ug/mL SEB overnight and the results are shown in the attached image. Before working into a multi-color panel, CD86-PE was titrated individually and stain index was calculated, indicating 1uL was the optimal volume. Since the recommended volume to use per sample from BD is 20uL it also very cost-effective since 1uL/sample looks great.
