Both M. tuberculosis and Zika virus presence/absence tests are two channel assays utilizing either cooperative primers or rapid probes. Cooperative primers double as PCR primers and hydrolysis probes, while rapid probes are innovated version of hairpin probes. Both rapid probes and cooperative primers are proprietary technology of Co-Diagnostics, Inc.Amplification of genetic material from either infectious agent is monitored in one channel, while amplification of internal positive control is monitored in another channel. The assay for detection of multiple drug resistance in M. tuberculosis is one-channel assay, based on amplification of several hyper-variable regions of M. tuberculosis genome in the presence of intercalating fluorescent dye and DNA probes. Subsequent high resolution melt reveals presence of resistance-conferring mutations within the interrogated regions.

We purchased the 4-channel MIC instrument and used it for development and verification of several of our commercial assays (as mentioned above in detail). MIC is extremely easy to setup, the software is intelligent, intuitive and possesses wide array of data analysis features. While most of our assays are real time PCR with hydrolysis probes, some of our tests also utilize high resolution melting of unlabeled probes or short amplicons. We were impressed by the excellent melt quality and very low noise, especially in the lower temperature regions, where most other instruments struggle. Temperature uniformity was excellent, as confirmed by consistent Ct values and tight clustering of the melt curves.
Another thing to highlight is the lack of need for calibration, otherwise an expensive process that has to be performed periodically.
