This review is for the GOBlot™ Western Blot Processor,  which has been discontinued and replaced by the GO-Blot-V2 Western Blot Processor.

We have so far bought one GOblot western processor and have used it with various antibodies, such as anti actin, GFP, Pgk1, and custom-made antibodies. The processor is very easy to set up, and it very reliably executes any of the chosen programs. The in-built programs provide sufficient choices for optimal time set-up for antibody incubation. We found that the western results showed nice signals with a low background. In fact, the background sometimes was even better as compared to westerns conducted the conventional way. We then proceeded to having two western membranes processed simultaneously, back-to-back, i.e. the protein sides of the membranes were facing outwards. Even though it appeared that the blocking solution barely covered the membranes, the western results were as good as when processing them individually! So far we have run the GOblot for about 50 times, and it is still working as if it was new. The processor is of satisfactory quality, and we would buy this product again without any hesitation.

Application

Western blotting

Starting Material

Western Blot Membrane

Protocol Overview

The manufacturer's protocol was used.

Tips

Add azide to the primary antibody solution to prevent contamination by bacteria.

Results Summary

The western results showed nice signals with a low background, which sometimes was better than that of westerns conducted the conventional way. We found that two western membranes can be processed simultaneously, back-to-back, giving results that were as good as when processing them individually.

Features Summary

Western processing involves many washing steps, and by automating these steps using this processor frees up your valuable time for other tasks/experiments.

Summary

The Good

Huge time saver, great results, easy to handle

The Bad

The retrieval of the primary antibody solution is not as complete as when doing westerns the conventional way (without machine).

The Bottom Line

The processor is of satisfactory quality, saves a huge amount of time, and we would buy this product again without any hesitation.
