We have used the Reichert SR7500DC for at least 5 years. In that time our application has been study the reconstitution and activity of proteins interacting with liposomes that were immobilized on the SPR surface. We have employed this system to study the binding of BCl-2 family proteins to liposomes and the removal of cholesterol from liposomes. In both cases the SR7500DC has permitted the quantitative study of the kinetics in a way that was not possible without the instrumentation. The reliability and stability of these measurements coupled with the programability for high-throughput assay development has been critical in allowing inhibitor and activator discovery and characterization. These experiments have immediate implications for translational application in the understanding and treatment of atherosclerosis and cancer therapeutics.
Binding of BCL-2 family proteins to liposomes.
Liposomes of defined composition and purified BAX, BID and BCLxl
The SR7500DC system was used to quantitatively the membrane binding, integration and protein--protein activation at the liposome surface.
The SR7500DC system was able to work at low, "physiologic" concentrations.
These studies identified the binding and integration steps of these proteins interaction with the membrane and how the integration was inhibited by cholesterol/
Stability, sensitivity and programability.
The SR7500DC system was a reliable workhorse.
Reliability, quantitative, programability and sensitivity
Cholesterol crystals can clog tubing
Best high through put surface plasmon resonance instrumentation value on the market that I have found.
