The overall goal of this research is to study where in the animal tissue the virus can go. The specific objective is to use luciferase reporter as a guide to hone in on a small pieces of the tissue that contain the infection. This product was chosen due to the availability within the department.

Protocol Overview

Tissue was washed well in PBS and imaged in IVIS to determine the background level of luminescence within the tissue. Tissue was next soaked in luciferin for 10 minutes on ice and was than incubated at 37 degrees C for 10 minutes prior to imaging in IVIS. The tissue containing foci of luminecence that were not present when tissue was imaged in PBS were excised and further studied.

Tips

The shift from cold to 37 degrees is likely beneficial

Results Summary

The tissue imaged pre luciferin contained minimal to no background luminecense. When tissue was exposed to luciferin foci of the infection were evident.
