The overall goal of this study was to phenotype the resident cells across various tissues. The more specific goal was to sort T cell sub types and perform RNA sequencing from the single cell. This product was chosen because the UV excitable fluorophore fit well into our panel.

Rhesus macaque PBMCs

Protocol Overview

PBMCs were isolated from blood and washed in PBS. Blue Fixable Live/Dead cell dye was added in 1:1000 dilution to approximately 1 million of cells for 15 minutes on ice protected from light.

Tips

Add dye before the samples were incubated with block buffer containing FBS

Results Summary

Using live/dead cell dye allowed us to select only for the viable cells and ensure that cells we sorted were worth experimenting with.

Additional Notes

Dye needs to be first resuspended in DMSO and should be stored at -20C
