The aim of this project is to study the role of the Wnt pathway in a mouse model of colon cancer. Beta-catenin is an important regulator in the Wnt pathway and in colon cancer it's expression is elevated. In the present experiment, beta-catenin expression was checked in a normal mouse colon tissue (pilot experiment).

Primary Incubation

1:200 dilution in blocking buffer; overnight at 4 deg Celsius

Blocking Agent

1x PBS + 1% BSA + 1% goat serum

Secondary Incubation

1:200 dilution (in 1xPBS + 1% BSA) of Goat-anti rabbit Alexa dye 594; 1 hour at r/t

Tertiary Incubation

DAPI was included in the last wash step

Detection

Results Summary

Beta-catenin expression was good enough to detect in a normal mouse colon tissue
