Anti-alpha Actin was to stain and detect smooth muscle cells harvested and grown from primary aortic tissue isolated from mice. Anti-alpha actin (sc-32251) resulted in a strong, characteristic staining pattern indicative of actin.

Sample

Aortic tissues from mice where treated in collagenous and expanded in cell culture for 3 days priory to cellular characterization with alpha actin.

Primary Incubation

A dilution of 1:50 anti-alpha actin in 2% DGS was incubated at room temperature for 2 hours.

Blocking Agent

10% Donor Goat Serum (DGS).

Secondary Incubation

A dilution of 1:100 of anti-Mouse-594 (Alexa series) in 2% DGS was incubated at room temperature for 1 hour.

Tertiary Incubation

N/A

Detection

594nm excitation on an inverted microscope.

Results Summary

The anti-alpha actin gave a very nice staining pattern of a large density of cells isolated from the mouse aorta. The staining pattern was indicative of the cytoskeletal protein actin.
