The long-term goal of this project is to test the effect of EGFR inhibition on the proliferative properties of human primary keratinocytes. The first step in this project was to determine the expression of EGFR by Western Blotting. This antibody from Cell Signaling Technology was used because more than 20 papers have cited this product.

Experimental Design and Results Summary

Applications

Western Blot

Sample

Human primary keratinocyte culture lysate

Primary Incubation

1:1000 in blocking buffer; 16 hr at 4 deg

Blocking Agent

5% BSA in 1xTBS-T(0.1%)

Secondary Incubation

Goat-anti Rabbit-IgG-HRP conjugated; 1:10,000 dilution in blocking agent

Tertiary Incubation

None

Detection

ECL regular

Results Summary

Human primary keratinocytes express good amounts of EGFR as detected by Western Blotting. 50 µg of total cell lysate was sufficient to give a good signal.
