This kit was used for quantifying Interleukin 6 in plasma samples of 21 days chickens for comparative analysis of immunological response. This kit can be used in other chicken body fluids. We selected this kit by suggestion of another researcher.

Starting Material

21 days chicken plasma

Protocol Overview

1 - Bring reagents and sample to room temperature;2 - Add 100 ul of standards and samples. Cover and incubate for 90 minutes at 37ºC;3 - Prepare Biotinylated antibody IL-6 Chicken 30 minutes before use; 4 - Wash plate 3X 2 min each;5 - Add 100 uL of Biotinylated antibody IL-6 Chicken. Cover and incubate for 60 minutes at 37ºC;6 - Prepare enzyme-conjugate liquid 30 minutes before use;7 - Wash plate 3X 2 min each;8 - Add 100 uL of Enzyme-conjugate liquid (except Blank wells). Cover and incubate for 30 minutes at 37ºC;9 - Wash plate 3X 2 minutes each.10 - Add 100 ul liquid Colour Reagent. Cover and incubate in the dark for up to 30 minutes at 37ºC;11 - Add 100 uL of Reagent C. Colour12 - Afetir Optica density (OD) at 450nm in at most 15 minutes after addition of Stop solution.

Tips

Read all the protocol before proceed and do not use diluted samples.

Results Summary

The standard curve has a detection range between 15,6 and 1000 pg/mL). The curve was drawn on a arithmetical scale, and the equation for quantyfing was obtained with a linear regression. The r² was iqual to 0,9967

Additional Notes

Sometimes, the mininum among of IL-6 can not be determinated even in undiluted samples
