Starting Material

21 days chicken plasma

Protocol Overview

1 - Place reagents and samples to room temperature 2 - Add 100 uL off standards and samples in the pre determined wells. Cover the plate and incubate for 60 minutes at 37ºC 3 - Remove the liquid and DO NOT WASH 4 - Add 100 uL of Detection Reagent A. Cover the plate and incubate for 1 h at 37ºC 5 - Wash the plate 5X of 2 minutes each; 6 - Add 100 uL of Detection Reagent B. Cover the plate and incubate for 1 h at 37ºC 7 - Wash the plate 5X of 2 minutes each; 8 - Add 90 uL of Substrate solution. Incubate for 20 minutes at 37ºC protected from the light. 9 - Add 50 uL of Stop solution. 10 - Measurment of the Optical density (DO) at 450nm;11 - build a standard curve with a regression line

Its appropriate standardize the method for the specific tissue before the real experiment.

Results Summary

The standard curve has a detection range between 7,8 and 500 pg/mL). The curve was drawn on a arithmetical scale, and the equation for quantyfing was obtained with a linear regression. The r² was iqual to 0,9884.

Sometimes, the mininum among of IL-1beta can not be determinated
