Protein Kinase D is family of Ser/Thr kinases that regulate multiple biological functions including cell migration, proliferation, apoptosis, transcriptional activation, angiogenesis, and Golgi organization and vesicle trafficking. In the present study, the molecular basis of Protein Kinase D recruitment to the Golgi complex was investigated. I had discovered that class I and Class II ARF proteins and more specifically ARF1 can function as a tethering factor for Protein Kinase D at the Golgi complex. These conclusions were based on protein-protein interaction assays, colocalization experiments, and Golgi functional assays after knockdown down of Protein Kinase D in cells. I had used PKD mu or PRKD1 antibody from Santa Cruz to detect the expression of this isoform in different cell lines.
Western Blot
Whole cell extract from HEK293T, HeLa, PANC-1, 3T3 and NRK cells
1:200 dilution at room temp for 1 hr
5% milk+0.1% Tween in 1xTBS
Donkey anti-rabbit IgG-HRP conjugated
None
Chemiluminescence
PRKD1 was found to expressed in the above mentioned cell lines and I had proceeded with the HeLa cell line for future experiments
It works but sometimes it was tricky to detect a signal
Failed few times to detect a signal
Not a bad antibody for PRKD1
