The RNase-Free DNase Set provides efficient on-column digestion of DNA during RNA purification using RNeasy® Kits. Generally, DNase digestion is not required for RNA purified with these kits, since silica-membrane technology efficiently removes the majority of the DNA without DNase treatment. However, complete DNA removal may be necessary for certain RNA applications that are sensitive to very small amounts of DNA.

Protocol Overview

Prepared the nucleic acid solution concentration according to the instruction. Added DNase I Buffer in the RNA sample. To this Added 1 μL DNase I (2 U) for up to 10 μg RNA in a 50 μL reaction, and incubate at 37°C for 30 minutes. Extract the RNA sample with phenol/chloroform to inactivate the DNase I. Then followed for RNA extraction.
