Efficient extraction of the total protein from the starting material is vital for success of any purification procedure. Complete disruption of cells and release of contents from cellular debris is the most important step in the process. Followed instructions for total protein extraction. After adding the rehydration buffer, samples were sonicated 3-4 times for cell lysis. This was centrifuged at 15000*g for 15 min. Then, I removed and transferred supernatant to another clean tube. This is used for immunoprecipitation.
Disruption by sonication is dependent by type of cells
I acquired a good yield of protein and it worked well for immunoprecipitation
