Goal of this research is finding specific anti-HA antibody. After transfection of HA-CAS to PC12 cells, specific anti-HA signal was detected in the cell nuclei of a few transfected cells.Thus, delivery of CAS gene, its expression and detection of HA tag in the expected location was successful

Leica confocal microscope
PC-12 cells were transfected ( by llipofectamine) with saCAS-HA tagged and 48 hours after transfection they were fixed with 4% PFA for 15 min, rinsed, permeabilised with 1% Triton for 15 minutes and stained with mouse anti-HA antibody (Covance #MMS-101R), dilution 1/1000 and custom rabbit anti-Sigma-1 receptor antibody for 2 hours. Secondary donkey-anti-rabbit Cy2 conjugated and donkey-anti mouse Cy3 conjugated antibody were applied for 1 hour. Cells were rinsed and counterstained with DAPI. Clear nuclear localization of saCAS is detected.
