We often use kit to purify the ChIP DNA after the reverse cross linking the DNA from binding. We prefer this kit because it's very easy to use and affordable price. The DNA quality from this procedure is very high, does not need any downstream purification for the sequencing.

We use chIP binding buffer 5 fold to the sample volume for the binding. The sample and binding buffer mixed at least 10 times with pipetman. The DNA and binding buffer complex was pass through column provided in the kit. Then column washed with wash buffer and eluted in 10ul volume of 10mM Tris- HCl. elution buffer.
Mix thoroughly with chIP binding buffer.
We obtain high quality DNA for sequencing. We elute samples in 10ul elution buffer gives in the range of 5-10ng/ul. The concentration varies depends on the interaction strength and antibody.
