I used QIAquick PCR purification kit to purify PCR amplified phoQ gene product of Salmonella enterica for cloning experiments. At the end I eluted in 20 microlter of nuclease free molecular biology grade water and got recovery of 80 %. This kit is very good to get rid off PCR components like dNTPs, enzyme and unused primers which otherwise interfere with cloning reaction. Quality of the purified product after purification was very good (260:280 ratio was 1.83).

Protocol Overview

I used 50 microliter of phoQ PCR product and recommended volume of binding buffer and allowed to stand at Room temperature for 10 minutes. Then followed the protocol as per the kit's instruction. Finally eluted DNA 20 microliter in the nuclease free molecular biology grade water.

Tips

Use nuclease free molecular biology grade water for final elution.

Results Summary

This kit performs very well and recovery of phoQ PCR product was 80% with good quality as checked by Nanodrop ND1000 spectrophotometer.
