The mouse mAb 610181 from BD Biosciences was used to stain E-Cadherin in human prostate cancer cells PC-3. The antibody produced good staining with non-permeabilized cells, consistent with the cell membrane localization of E-cadherin.
Immunofluorescence
Human prostate cancer PC-3 cells, grown on glass coverslips, fixed in 1% formaldehyde for 20 min RT, and permeabilized in 0.2% Triton X-100 for 5 min, RT.
Overnight, at 4-8 degrees Celsius, at 25 ug/mL in PBS/1% BSA
10% normal goat serum; 1 hr; RT
Goat anti-mouse-DyLight488; 1:1,000 in PBS/1%BSA
NuncBlue was used to highlight cell nuclei
IF microscope; Olympus
This antibody produced good results in E-cadherin-positive human tumor cells fixed and permeabilized with Triton X-100.
