In our lab we use this kit combined with MACS gentle dissociator to dissociate the adult brain from the mouse. The protocol is highly optimized by Miltenyi Biotec for mouse brain that yields high amount single cells from the mouse brain. We use routinely in our lab that yield consistent reproducible result across different hands.
To obtain single cells from adult mouse brain
Brain tissue
The MACS gentle dissociator is loaded with pre-programmed that optimized for Brain dissociation. We used 400mg of mouse brain tissue per protocol. The brain was dissociated by both mechanical (MACS dissociator) and enzymatic methods (supplied with kit).
Don't exceed the amount of tissue per experiment
We consistently obtained individual single cells from brain tissue and yield was close million cells.
