Measurement of IL-8 in media supernatant is a commonly used technique in immunology labs. It is secreted by a wide variety of cells, and can be utilized as a control to measure the cell's response to various stimuli. ELISA kits can be expensive, but this option from BioLegend offers the best of both worlds: high quality kit, and very reasonable pricing!

The capture antibody is added to the wells overnight, then non-specific binding is blocked before you add the samples and standards. A detection step follows, and measurement is carried out on a plate reader. Overall, it takes a whole day, but with long breaks in between!
Washing steps are critical!
The standard (provided) produces excellent data, which gives you confidence in the results you get from your samples. The sensitivity is excellent (15.6-1000pg/ml).
All ancillary reagents can be made in house. No need to buy any extra stuff!
