Anti-CD31 was used to stain human synovial cells to identify the presence of endothelial cells

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Human synovial cells

Primary Incubation

30 minute incubation at 4oC. Live/dead stain.

Blocking Agent

No specific blocking step. 2% FCS in staining buffer.

Secondary Incubation

30 minute incubation at 4oC. Antibody cocktail used at this point containing antibody at 3ug/ml.

Tertiary Incubation

None

Detection

BD LSR X-20 Fortessa. Analysis using FlowJo v10

Results Summary

Antibody worked well identifying a CD31 positive population. Staining intensity was moderate but allowed the identification of a small number of CD31 positive cells.

Summary

The Good

Antibody worked well to identify positive cells.

The Bad

The staining intensity was moderate.

The Bottom Line
