Gene expression of G6Pase (Glucose 6 Phospatase) was studied in liver samples. Total RNA was isolated. RNA is then converted into cDNA which is amplified by SYBR Green PCR Master Mix by following the kit protocol.

Experimental Design and Results Summary

Application

Gene expression analysis

Starting Material

Rat liver sample

Protocol Overview

Rat liver was homogenized in TRIZOL reagent and total RNA was isolated by TRIZOL method. The isolated RNA was then converted into cDNA. cDNA template was amplified by adding primers and SYBR Green PCR Master Mix according to the kit instructions and amplified in Real-Time PCR machine. Contents were mixed properly. Amplification parameters were also followed from the kit. The SYBR Green dye binds with the double stranded DNA during amplification and fluorescence was detected.

Tips

Kit should be stored at 2 to 8°C

Results Summary

The amplification was done Real-Time PCR machine and amplified products were observed in 3% agarose gel.
