Aim of the study was to test the effect of Gα(q) specific siRNA on Gα(q) expression levels in human smooth muscle cells. Cells which were cultured in complete medium and transfected with Gα(q) siRNA using lipofactamine 2000. After 50 hrs of transfection, cells were washed in cold PBS, protein extracts were prepared, resolved in 10% SDS-PAGE and immunoblotted for Gα(q) protein levels using Cell Signaling Technology Gα(q) antibody. Selection of antibody was based on information provided in antibody flyer.

Results Summary

Cell Signaling Technology Gα(q) antibody worked very well and detected only one highly specific, bright band of Gα(q). However, Gα(q) siRNA knockdown its target protein with high efficiency.
