The aim of the study was to test the time course effect of growth factor on Gα13 interaction (association/disassociation) with growth factor specific G-protein coupled receptor (GPCR) in mammalian smooth muscle cells. Cells were time dependently treated with growth factor, proteins were extracted, equal amount of proteins were co- immunoprecipitated with receptor antibody. Co-immunoprecipitated cell lysates were incubated with specific protein A/G beads, immunocomplexes were resolved in 10% SDS-PAGE, and immunoblotted with anti- Gα13. Selection of antibody was based on compatibility of antibodies for immunoprecipitation information as provided in antibody flyer.

Gα13 antibody from SantaCruz biotech has detected a specific band of Gα13 at nearly molecular weight of 40 kDa. However, time dependent disassociation of Gα13 from growth factor receptor was observed.
