We needed a LAMA1 antibody to validate some microarray data that we had involving our human Y79 retinoblastoma cells. We chose this antibody because it was polyclonal and would be compatible with other mouse antibodies for double immunofluorescence.

Sample

Cytospin of Y79 human retinoblastoma cells

Primary Incubation

One hour at room temperature with the LAMA1 antibody at 10 ug/ml in PBS with 0.05% Tween20

Blocking Agent

None

Secondary Incubation

45 minutes at room temperature with anti-Rabbit Dylight 488 (Vector D1148) at 5 ug/ml

Tertiary Incubation

Vectashield mounting medium containing DAPI counterstain

Detection

Fluorescence microscopy- Nikon microscope with SPOT camera

Results Summary

The LAMA1 antibody provided excellent results as seen in the accompanying figure. The level of background fluorescence was within acceptable limits.
