CD56 is expressed on the vast majority of resting and activated natural killer (NK) cells, as well as on a minor subset of CD3+ T cells. Our lab routinely uses this antibody to identify NK cells in peripheral blood samples. The product gives excellent results, making it very easy to distinguish between CD56+bright and CD56+dim populations.

Human peripheral blood mononuclear cells isolated from whole blood by standard Ficoll density gradient separation

Primary Incubation

PE-conjugated anti-human CD56 antibody (Miltenyi 130-090-755), incubated with approximately 1x10^6 cells for 1 hour on ice at 1:10 dilution in a total volume of 100ul PBS+2% FCS, 1mM EDTA, 0.1% sodium azide.

Results Summary

This antibody worked excellent in FACS analysis of human PBMC. As expected, two populations of CD56-expressing cells were identified, one with high expression (bright) and another with lower expression (dim). Very little background staining compared to unlabelled cells was observed.

Additional Notes

For flow cytometric analysis, gating was performed on the lymphocyte population. Monocytes were negative (not shown).
