NFATc1 is an important transcription factor that is upregulated during differentiation of macrophages into osteoclast. NFATc1 is necessary and required for this process and is commonly used as a marker of differentiation. Here we test an antibody from Santa Cruz against murine NFATc1 in cells in different steps of the OC differentiation process and show high specificity and sensitivity to our cellular lysates.

Results Summary

We used WT or KO murine bone marrow macrophages deficient in their ability to differentiate into osteoclasts and either retrovirally overexpressed an empty vector or NFATc1 construct to rescue the KO's differentiation defect. As observed, in bone marrow macrophages, NFATc1 expression in empty vector cells are low, as expected. With NFATc1 overexpression in macrophages, there is increased expression of this protein. As the WT cells differentiate into mature osteoclasts, in empty-vector samples, we see native upregulation of NFATc1, and blunted upregulated in the KOs. However NFATc1 overexpression is strongly exhibited in both WT and KO osteoclasts. 10 ug of protein was lysed in RIPA buffer per lane.
