This product was used to measure cytokine levels in lung homogenate. It was easy to use, we use a lot of their ELISAs and always have good results.

Starting Material

Murine lung homogenate supernatant.

Protocol Overview

Lungs from Mtb infected mice were homogenized in 5mL PBS-T. Homogenate was aliquoted and spun down at max speed, and the supernatant was filtered through a 0.2um PVDF filter. Sample was diluted 1:3 in the appropriate buffer as per product guidelines.

Tips

May need to dilute sample to get within the standard curve.

Results Summary

This was easy to use, we use a lot of their ELISAs and always have good results. We went through several of these kits, and at one point, they switched the detection Ab for the IFNG kit (I think they went from a polyclonal to a monoclonal)- and didn't explicitly say that in the product info. I saw some variations in my results, and wasn't really happy about that, but once I got in touch with technical service they sent me a replacement kit and extra Ab from each lot so I could specifically compare the different Abs. Ultimately, the two kits performed similarly and I appreciated the technical service agent working with me on this - so I will definitely stick with their company in the future.

Summary

The Good

Easy to use - good technical service.

The Bad

Had some variation between a kit, but I think it was just a bad kit. Customer service replaced the kit for free and since then haven't had any problems.
