This kit provides clear instructions and good yield (~90%) of microglia purification with CD11b+ selection.
Microglia purification
Dissociated murine CNS tissue after Percoll gradient
Make single cell suspension of CNS tissue with enzyme digestion/myelin separation with Percoll.
Make sure the MACS buffer is consistently cold!
Cells from the purification were counted and plated; the remaining cells were used for flow cytometry verification of purity, which was ~90%. Additional columns could have been used for increased purity.
