We recommend using the antibody rather concentrated. A dilution of 1:2000 for WB did not give good results but using it 1:200 works fine. We also tested the antibody on paraffin-embedded brain tissue sections. The antibody works also for IHC-P stainings. Use a 1:500 or even a higher dilution otherwise there will be a lot of background.
Immunohistochemistry
C57BL/6 mice GBM-bearing brain
1:200 dilution of primary antibody; incubation: overnight at 4°C
3% H2O2 for 20 min at RT, 1%BSA 5% NHS for up to 1 hour at RT
Antibody diluted in goat serum; dilution according to manufacturer's manual (Vectastain ABC system); incubation: 1 hour at RT
Vectastain ABC system; dilution according to manufacturer's manual; 30 min at RT
Dako DAB Substrate Detection System
Good results. The tested antibody dilutions 1:50 and 1:100 show a high tissue background. 1:200 gives better results with almost no background. One can test also dilutions greater than 1:200.
