The general aim was to measure cytokine release, especially the release of IL-1beta in the BV2 stimulated cells. Stimulation is done to mimic the stress response.
Measurement of IL 1 beta in BV2 cells

Coat overnight with capture antibody. Block the plate after 24h, add your supernatants and detect the antibody. After this step, add the enzyme and the substrate. Stop with stop solution and read the plate.
Wash more times to enhance effectiveness
I got higher values of cytokine release in my cells which have been stimulated with higher endotoxins.
