The aim of this study was to develop a panel that allows us to distinguish the general surface markers on human blood. With CD19-PE/Cy7 we could identify the B-cells in human blood well.

Primary Incubation

1.) 80 µl EDTA blood plus 20 µl antibody cocktail (in FACS Buffer) was incubated for 30 min at RT. 2.) Lysis of erythrocytes was achieved by adding 1 ml of Lysis Buffer and incubating for 10 min at RT. 3.) Washed two times with PBS; centrifuged at 400 x g for 5 min, trash supernatant. 4.) Resuspended cells in 200-300 µl FACS Buffer, FACS analysis.

Results Summary

You get a nice distinction between B-cells and T-cells. The antibody is used with a final dilution of 1:500.
